First paper on specimen preparation for X-ray fluorescence microtomography by femtosecond laser sectioning

The research group of Christoph Fahrni (Georgia Institute of Technology, Atlanta, GA, USA) and co-workers has published high resolution 3D visualization of transition metals in a complete zebrafish embryo using femtosecond laser for sample preparation. Transition metal homeostasis is pivotal for many biological processes and disregulation may lead to metal overload or deficiency which is regarded to play a role in several diseases, e.g. Parkinson, Alzheimer. To understand the underlying mechanism of transition metal homeostasis , it is essential to analyse metal distribution in cells, tissue or whole organisms using modern microanalytical techniques. X-ray microtomography requires sufficient mechanical sample stability for scanning on a rotational stage, which is critical when using soft tissue specimen. With zebrafish embryo as a test sample the group of C. Fahrni was able to acquire a complete set of tomographic projections and reconstruct the 3D-distribution of several metals while the structural integrity of the sample was preserved by using the TissueSurgeon for sample preparation.
